چکیده

Streptomycin, an aminoglycoside, binds to and modifies prokaryotic ribosomes leading to a decrease in protein synthesis and the production of mistranslated proteins. Previous studies reported that Escherichia coli B23, under sub-lethal concentrations of streptomycin, exhibited an increase in capsule polysaccharide (CPS). The mechanism through which this increase in CPS occurs was unknown. In this investigation, we hypothesized that the increase in CPS in Escherichia coli B23 correlated with the increase in mistranslated protein and not simply due to a decrease in protein synthesis. Mistranslation in protein was measured by changes in the Michaelis constant (KM) and maximum reaction rate (Vmax) of induced βgalactosidase while CPS production and protein synthesis was measured using an anthrone assay and optical density respectively. Our results are inconclusive on the correlation between protein mistranslation and CPS production but indicate that decreased protein synthesis does not consistently correlate with an increase in CPS.

The Effect of Sub-Inhibitory Streptomycin on Capsular Polysaccharide Production and Streptomycin Resistance in Escherichia coli

Previous studies have shown that pre-treatment of Escherichia coli B23 with subinhibitory concentrations of aminoglycosides leads to a subsequent increase in the level of capsular polysaccharide, as well as an increased resistance to streptomycin. Using capsular polysaccharide synthesis pathway mutants, we investigated the correlation between the level of capsular polysaccharide and streptomyci...

Expression of E.coli capsular polysaccharide requires the KfiB protein:A Structural based analysis

Abstract Background and objectives: important virulence factor for many invasive bacterial pathogens of humans. Escherichia coli offer a model system to study the mechanisms by which capsular polysaccharides are synthesized and exported onto the cell surface of bacteria. Biosynthesis of the E consists of the repeat structure -4) GlcA- (1, 4)-GlcNAc- (1-, requires the KfiA,...

Prokaryotic expression, purification and immunogenicity analysis of CpsD protein from Streptococcus iniae

Streptococcus iniae is a major cause of serious bacterial infections in both fish and human beings. Capsular polysaccharide (CPS) of S. iniae is vital to evade phagocytic clearance of the host and serves as an important protective antigen of S. iniae infection in aquatic animals. The CpsD gene was determined to be highly conservative in capsule polysaccharide operon. Prokaryotic expression of t...

Effects of streptomycin and kanamycin on the production of capsular polysaccharides in Escherichia coli B23 cells

Aminoglycosides are a class of antibiotics that inhibit protein synthesis in bacteria by binding to the 30S ribosomal subunit. However, it has been reported that certain aminoglycosides have secondary effects on the cell’s overall carbohydrate concentration. Previous studies have also reported an increase in capsule size. This study attempted to investigate the effects of kanamycin and streptom...
